Organization: The Department of Biochemistry and Molecular Biology has an opportunity available for a Postdoctoral Associate. Dr. Feng-Chun Yang’s laboratory is focusing on understanding the cellular and molecular mechanisms mediating pathogenic development of hematopoietic malignancies.
Project information: Asxl1 gene is mutated and/or deleted with high frequencies in multiple forms of myeloid malignancies including MDS, MPN and AML. The majority of the ASXL1 mutations in these patients are heterozygous, leading to nonsense/frameshift, suggesting loss of function. Mutations in ASXL1 are associated with poor prognosis. We recently found that normal interaction of Asxl1 and BAP1 is required for the hematopoiesis. One of our projects is to clarify whether disruption of Asxl1/Bap1 interaction leads to myeloid malignancies. Candidate will utilize murine models of Asxl1, Bap1 to characterize the phenotypes in various Asxl1- Bap1 deficient mice, including Asxl1+/- and Bap1f/f;MxCre or Vav1Cre mice. In addition, the candidate will study the molecular mechanisms by which Asxl1-Bap1 interaction regulates normal hematopoiesis and exerts their tumor suppressor function in myelopoiesis. Furthermore, ChIP-Seq will be used to identify the differential genes targeted by Asxl1 and Bap1. Dr. Li will analyze Asxl1- and Bap1- associated histone H3 methylation and dissect co-deletion of Asxl1 and Bap1 induced misregulation of H3 methylation in HSCs.
Qualifications / requirements: Minimum qualifications include a PhD in a science related field. No experience necessary. Any appropriate combination of education, certifications, and/or relevant work experience may be considered.
